反胶束萃取蛋白质中静电相互作用能的研究

Research on Electrostatic Interaction Energy Related to Txtraction of Protein by Reversed Micelles

  • 摘要: 在双球坐标系中,求解了描述蛋白质位于反胶束水池中偏心位置时所形成扩散双电层的非线性Poisson-Boltzmann方程,同时也求解了蛋白质与反胶束单独存在时,以及蛋白质位于反胶束水池中心位置时所形成扩散双电层的上述方程。求解时,在反胶束内表面处使用了非线性的电荷调节边界条件。基于上述数值解,应用自由能方法,计算了蛋白质被萃取进入反胶束水池过程中的静电相互作用能量,在此基础上,就反胶束水池中、耦合水相中离子种类、离子强度、被萃取物蛋白质分子大小与电荷量、以及偏心距对萃取体系的效果进行了预言。

     

    Abstract: The non-linear Poisson-Boltzmann equation (PBE) which describes the electrical double layer established when protein is situated eccentrically in the water wool of reversed micelles is solved numerically in bispherical coordinates. The PBE which describes the electrical double layer established when protein and reversed micelles exist separately, protein is situated in the water wool of reversed micelles concentrically, are also solved numerically, the non-linear charge regulation boundary condition has been treated on the inner surface of reversed micelles. Based on the above numerical solution, a calculation is performed for the electrostatic interaction energy related to the eatraction of protein into the water wool of reversed micelles by free energy method. The effects of ion species, concentration in the water wool of reversed micelles and coupling aqueous phase, molecule size and number of charge of protein extracted, eccentric distance on the extraction system have been predicted based on the results of electrostatic interaction energy calculation.
